The microscope reticle is a small piece of glass that has been etched with a grid or a ruler that either fits into your microscope eyepiece as an accessory or that comes integrated into your ocular. This grid or ruler is then visible when you look through your eyepiece, making it easier to make measurements when observing or interacting with a specimen sample on your microscope stage.
Taking accurate measurements from a microscope reticle requires calibration, most often through the use of a stage micrometer. This measurement tool is designed to be placed on an otherwise empty microscope stage and then observed with your reticle-equipped eyepiece. Positioning one end of the reticle to one end of the micrometer provides you with a sense of scale, allowing you to note what types of units you should be using while measuring your sample. If you switch to an objective lens with a different magnification, you’ll need to recalibrate your microscope reticle all over again.
